Abstract Background and Aims Acute tubular injury accounts for the most common intrinsic cause acute kidney (AKI). The scattered cell (STC) phenotype was discovered as a uniform reaction of tubule cells triggered by injury. Our group first to identify an inducible transgenic mouse (PEC-rtTA-mouse) specifically labeling STCs with eGFP. Analysis transcriptional factors associated signaling pathways might reveal function role in AKI. Method Here, we performed single-cell RNA sequencing unilateral ischemia-reperfusion murine model AKI 8, 24, 48 hours 6 12 days after induction. Results Genes expressing proximal proteins transporters were markedly downregulated during transition into STC upon injury; but expression recovered over time resolution re-differentiated cells. This provides evidence that is transient reversible Among phenotype, could 2 sub-clusters; highly proliferating sub-cluster cycle analysis showed highest proportion cycling second eGFP-positive cluster appeared very early expressed distinct set genes (defined 7 anchor genes).
